Patterns of avb3 Expression in Primary and Metastatic Human Breast Cancer as Shown by 18F-Galacto-RGD PET

چکیده
The integrin avb3 is a key player in angiogenesis and metastasis. Our aim was to study the uptake patterns of the avb3-selective PET tracer 18F-galacto-RGD in invasive ductal breast cancer. Methods: Sixteen patients with primary (n 5 12) or metastasized breast cancer (n 5 4) were examined with 18F-galacto-RGD PET. Standardized uptake values (SUVs) were derived by region-ofinterest analysis, and immunohistochemistry of avb3 expression was performed (n5 5). Results: 18F-Galacto-RGD PET identified all invasive carcinomas, with SUVs from 1.4 to 8.7 (mean 6 SD, 3.6 6 1.8; tumor-to-blood and tumor-to-muscle ratios, 2.7 6 1.6 and 6.2 6 2.2, respectively). Lymph-node metastases were detected in 3 of 8 patients (mean SUV, 3.36 0.8). SUVs in distant metastases were heterogeneous (2.96 1.4). Immunohistochemistry confirmed avb3 expression predominantly on microvessels (5/5) and, to a lesser extent, on tumor cells (3/5). Conclusion: Our results suggest generally elevated and highly variable avb3 expression in human breast cancer lesions. Consequently, further imaging studies with 18F-galacto-RGD PET in breast cancer patients for assessment of angiogenesis or planning of avb3-targeted therapies are promising.

Noninvasive Visualization of the Activated αvβ3 Integrin in Cancer Patients by Positron Emission Tomography and [18F]Galacto-RGD
BACKGROUND The integrin alphavbeta3 plays an important role in angiogenesis and tumor cell metastasis, and is currently being evaluated as a target for new therapeutic approaches. Several techniques are being studied to enable noninvasive determination of alphavbeta3 expression. We developed [(18)F]Galacto-RGD, a (18)F-labeled glycosylated alphavbeta3 antagonist, allowing monitoring of alphavbe...
متن کاملNon-invasive assessment of inter-and intrapatient variability of integrin expression in metastasized prostate cancer by PET
PURPOSE Due to the high expression of the integrin αvβ3 not only on endothelial cells, but also on mature osteoclasts and prostate cancer cells, imaging of osseous metastases with αvβ3-targeted tracers seems promising. However, little is known about the patterns of αvβ3-expression in metastasized prostate cancer lesions in-vivo. Thus we evaluated the uptake of the αvβ3-specific PET tracer [18F]...
متن کاملmicroPET of Tumor Integrin avb3 Expression Using 18F-Labeled PEGylated Tetrameric RGD Peptide (18F-FPRGD4)
In vivo imaging of avb3 expression has important diagnostic and therapeutic applications. Multimeric cyclic RGD peptides are capable of improving the integrin avb3–binding affinity due to the polyvalency effect. Here we report an example of 18F-labeled tetrameric RGD peptide for PET of avb3 expression in both xenograft and spontaneous tumor models. Methods: The tetrameric RGD peptide EfE[c(RGDy...

Objective(s): We performed a phase IIa clinical trial of trans-1-amino-3-18Ffluoro-cyclobutane carboxylic acid (anti-18F-FACBC), a synthetic amino acid analog for PET, in patients with metastatic prostate cancer. Methods: The study subjects consisted of 10 untreated prostate cancer patients having lymph node and/or bone metastasis. Five patients underwent whole-body PET 5 and 30 min after intra...
متن کاملComparison of cyclic RGD peptides for αvβ3 integrin detection in a rat model of myocardial infarction
BACKGROUND Expression of αvβ3 integrin is increased after myocardial infarction as part of the repair process. Increased expression of αvβ3 has been shown by molecular imaging with 18F-galacto-RGD in a rat model. The 68Ga-labelled RGD compounds 68Ga-NODAGA-RGD and 68Ga-TRAP(RGD)3 have high specificity and affinity, and may therefore serve as alternatives of 18F-galacto-RGD for integrin imaging....
